摘 要:目的:研究陇中损伤散对兔激素性股骨头坏死模型的治疗效果及其作用机制。方法:成年青紫蓝兔75只,随机分为6组:正常组(N)、模型组(M)、健骨生丸组(Y)、陇中损伤散高、中、低剂量组(SH组、SM组、SZ组)。采用内毒素加激素诱导的方法建立早期激素性股骨头坏死模型。造模后2周,各组动物给予相应的药物或生理盐水灌胃。用药后2、4、8周心脏采血,检测血浆中SOD活力与MDA含量的变化。结果:SH组、SM组和SL组第8周血浆总SOD活力提高,MDA含量下降;其中SH组和SM组SOD活力与M组比较差异有统计学意义(P<0.05),SL组SOD活力与M组比较差异无统计学意义(P>0.05);SH组、SM组和SL组MDA含量与M组比较差异有统计学意义(P<0.01)。用药后第2到8周,各检测指标M组与N组比较差异有统计学意义(P<0.05或P<0.01),Y组与M组比较差异也有统计学意义(P<0.05或P<0.01)。结论:陇中损伤散具有治疗早期激素性股骨头坏死的作用,其机理可能是清除自由基,部分改善血管内皮功能受损状态,改善微循环,降低骨内压。Objective : To study the therapeutic efficacy and mechanism of action on Longzhong-Sunshangsan ( LZSSS, a TCM composition) to the models with steroid induced avascular necrosis of femoral head (SANFH) in rabbits. Method :75 rabbits were randomly divided into 6 groups : normal control group ( group N), model group ( group M), JianGuSheng pills group ( group Y) and three-dose LZSSS groups (groups of SH, SM and SL). Rabbit models of SANFH in earlier period were established by injecting with endotoxin and methyl- prednisolone. After 2 weeks, all rabbits were given medicines and Saline via gastrogavage respectively. Blood samples were collected from heart puncture on times of 2 weeks, 4 weeks and 8 weeks after treating. The indexes of blood rheology MDA and SOD were measured. Re- suits : The plasma SOD activities of SH, SM and SL groups were significantly ascended ( P 〈 O. 05 ) , and the plasma MDA level was obviously descended ( P 〈 0. 01 ). On SOD activities, there had significant deviation between group M and group SH as well as group SM ( P 〈 0. 05) , but had no between group SL and group M (P 〉 0. 05). On MDA levels, there had significant difference between groups of SH, SM and SL with group M (P 〈 0. 01 ). From 2 weeks to 8 weeks after treating, there had significant difference on all indexes of detections in group M and group N as well as group Y and group M (P 〈 0. 05 or P 〈 0. Ol ). Conclusion: LZSSS had the function of treating SANFH. It' s mechanism might that it could clear away oxygen free radical, protect partil damaged states of emlothelial cells, improve microcirculation of femoral head and decrease intramedullary pressure.
