摘  要:目的确定二苯乙烯苷含药血清作用于老年性痴呆(AD)细胞模型的最佳条件。方法利用细胞计数、MTT方法确立β淀粉样蛋白(Aβ)25-35片段在建立AD细胞模型时的浓度及筛选二苯乙烯苷含药血清作用的最佳浓度,分别观察不同浓度血清及不同浓度Aβ25-35对AD细胞模型存活率的影响。结果各种血清以5%浓度组的细胞存活率最高。在5%血清浓度下,Aβ25-35 0μmol/L和5μmol/L浓度组中各含药血清组细胞存活率均明显高于正常大鼠血清组(P<0.05或P<0.01),但在10μmol/L Aβ25-35浓度时差异无显著性。结论选用5μmol/L Aβ25-35及5%血清浓度建立NG108-15 AD细胞模型及药效考察体系较为合适,在此条件下能客观地评价二苯乙烯苷含药血清的药效。Objective To confirm the optimum conditions of TSG- containing serum affect on cell model of Alzheimer's disease (AD). Methods Cell counting and MTT technique were used to determine the optimum concentration of amyloid β25 - 35 (Aβ25 - 35) and drug - containing serum in establishing cell model of AD, then observed the cell survival rate of the cell model after treated with different concentration of rat's serum and of amyloid β25 - 35, respectively. kinds of serum. At the Results 5% concentration of serum resulted in the highest cell survival rate among all 5 % concentration of serum condition, the cell survival rate of drug- containing serum groups were higher than the normal serum group at 0μmol/L and 5μmol/L concentration of Aβ25 - 35 ( P 〈0.05 or P 〈0 01 ), but had no difference at 10μoml/L concentration Conclusions It is the optimum condition to select 5μmol/ L concentration of Aβ25 - 35 and 5 % concentration of drug - containing serum to establish the NG108 - 15 cell model of AD and the best system to assess the drug efficacy. Under these conditions can he obiectively assessed the drug efficacy of TSG- containing serum.
