摘  要:本文报道了用小鼠胸腺细胞MTT比色分析法检测淫羊藿多糖、猪苓多糖与羧甲基茯苓多糖及微量元素锌对正常人外周血单个核细胞产生白细胞介素2(IL_2)的影响,结果表明,淫羊藿多糖在100μg/ml和1000μg/ml剂量对人IL_2的产生有显著的增强作用,在2500μg/ml和5000μg/ml表现出抑制作用;猪苓多糖与羧甲基茯苓多糖合用在一定浓度下也有显著的增强作用;微量元素锌在0.153~765μmol/L均有显著的增强作用。并证明,各药对IL_2产生的作用并非药物的直接作用所致,淫羊藿多糖高浓度的抑制作用亦非该药的细胞毒作用所致。本文建立了用小鼠胸腺细胞做靶细胞的MTT比色分析法并与~3H—TdR掺入法进行了比较,获得较好的一致性。The effects of Epimedium Polysaccharide(EPS), Grifola Polysacchari de (GPS), Carboxymethyl-Pachmaran(CMP) and Microelement zinc on IL_2 production of normal human peripheral blood mononuclear cells(PBMC) were reported in the paper. IL_2 activity was assayed by MTT colorimetric assay using thymocytes of mice as target cells. The results showed that IL_2 production was remarkbly increased by 100μg/ml, 1000μg/ml of EPS and was significantly inhibited by 2500μg/ml, 5000μg/ml of EPS The significant augmentation on human IL_2 production was manifested when mixture of GPS with CMP of certain concentration. The zinc of 0.153~765μ mol/L concentrations possessed significant incresing effect.The present study shows that increase of every drugs on IL_2 production was not directly effect of drugs inhibiting effet of EPS of high concentration was not also cytotoxic effect. The MTT colorimetric assay using thymocytes of mice as target cells was established and compared with 3~H-TdR incorporation assay. It showed that the results determined by MTT colorimetric assay were incomformity with those obtained by 3~H-TdR incorporation assay.
