摘  要:目的探讨Homer与神经元缺血性损伤中谷氨酸(Glu)、γ-氨基丁酸(GABA)相互关系。方法培养大鼠脑皮层原代神经元,先分为对照组、空载体组和Homerla siRNA转染组,检测Homerla变化。再将上述三组各分成正常组,缺氧缺血再灌注损伤后30min、1h、3h、12h、24h、48h共7小组,并进行Glu、GABA和Homerla的检测。结果①转染组神经元Homerla表达量明显减少;②损伤中,乳酸脱氢酶(LDH)活性增强;与其它两组比较,转染组培养液LDH活性最低;③损伤中,Glu和GABA与LDH变化正相关;Glu和Homerla的变化成反比;而GABA和Homerla的变化成正比。结论缺血性神经元损伤后,Homerla蛋白不仅调控Glu变化,还通过调控GABA变化保护神经元,在兴奋性和抑制性神经递质平衡中发挥重要作用。Objective To investigate the relationship between Homer protein and glutankate (Glu) and gamma-aminobutyric acid (GABA) associated with hypoxic-ischemic damage in cultured neurons. Methods Cortical neurons were cultured in vitro. RNA interference technology was performed to knock down Homerla gene, and the expression of Homerla in control group, empty vector groups and Homerla siRNA groups were examined. Then the neurons were divided into seven subgroups: normal control group and post-injury groups (30 min group, 1 h group, 3 h group, 6 h group, 24 h group and 48 h group). Results Compared with control group, the expression of Homerla was inhibited after siRNA transfection. Lactic dehydrogenase (LDH) concentrations increased greatly after hypoxic-ischemic damage. There was a positive correlation among concentration of Glu, GABA and LDH. But there was a negative relationship between Glu concentration and the expression of Homerla. There was a positive relationship between GABA concentration and the expression of Homerla. Conclusion After hypoxic-ischemic neuronal damage, Homerla not ouly modulates Glu but also regulates GABA to protect against hypoxic-ischemic neuronal damage. Homer protein plays an important role in the balance between excitatory and inhibitory neurotransmitter.
