摘  要:为了解葛根素对体外培养的人腹膜间皮细胞氧化应激反应的影响,取择期手术患者的网膜间皮细胞体外培养,第三代细胞用于实验。细胞同步后分5组观察,对照组加入等量的RPMI1640与完全培养基,PDS组加入等量的腹膜透析液(PDS)与完全培养基,A、B、C组均为加入等量的含葛根素的PDS与完全培养基,葛根素终浓度分别为50μg/ml、100μg/ml、200μg/ml。细胞培养48 h后,用比色的方法检测细胞上清液中氧化应激指标丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)浓度并比较。结果显示腹膜间皮细胞在PDS干预下,上清液MDA含量较对照组显著升高,SOD水平和GSH水平较对照组显著降低。添加葛根素再干预的A、B、C组MDA水平都显著低于PDS组,SOD水平和GSH水平都显著高于PDS组。实验表明葛根素具有抑制葡萄糖PDS对腹膜间皮细胞氧化应激反应的作用。To investigate the effect of Puerarin on the oxidative stress in cultured human peritoneal mesothelial cell. human peritoneal mesothelial cells oltained from patients who underwent surgical operation were cultured in vitro. At first, cells from the third passage were incubated with RPM I1640 culture medium containing 0. 1% FBS for 24 hours. Then, they were divided into five groups (RPMI1640 culture medium as control, PDS containing 2. 5 % glucose as PDS group, and PDS containing 2.5 % glucose and 100μg/ml, 200μg/ml or 400μg/ml Puerarin as group A, group B and group C). Each group was supplemented with equal volume of RPMI1640 culture medium containing 20% FBS. After 48 h incubating, MDA, SOD, and GSH in supernatant of the cell culture medium were detected and compared. Significantly lower level of MDA in supernatant was found in control group(P〈0.01), as well as in groups A, B and C (all P〈0.05), as compared with that in PDS group. And, higher level of SOD and GSH were in control group(P〈0.01) and groups A, B and C(all P〈0.05), than those in PDS group. Conclusion: Puerarin can reduce the oxidative stress induced by commercial PDS in cultured human peritoheal mesothelial cell.
