摘  要:目的观察尾加压素Ⅱ(UrotensinⅡ,UⅡ)对大鼠肾间质成纤维细胞(RFB)的促增殖作用,探讨UⅡ在肾纤维化发生发展中的作用。方法体外培养大鼠肾成纤维细胞并鉴定,采用MTT法、流式细胞仪(FCM)法分别观察不同浓度UⅡ作用下,大鼠肾成纤维细胞数、细胞周期的变化。结果UⅡ可以促进RFB的增殖活性,随着UⅡ作用浓度的增加,MTT吸光度值呈先升高后降低的趋势,其中1×10-9和1×10-8mol/L UⅡ组作用显著(P<0.05);在1×10-10,1×10-9,1×10-8,1×10-7mol/L UⅡ作用下,RFB S期细胞百分率均较对照组明显增加,而G0-G1期细胞百分率均较对照组明显降低(P<0.01);在1×10-7mol/L UⅡ作用下,上述各参数与对照组比较无统计学意义(P>0.05)。结论UⅡ能促进大鼠肾成纤维细胞增殖,影响细胞周期,这可能提示UⅡ在肾纤维化发生和发展过程中发挥着重要作用。Objective To observe the effects of urotensin Ⅱ ( U Ⅱ ) on the proliferation of the renal interstitial fibroblasts(RFB) of rats and explore the role of U Ⅱ in the renal interstitial fibrosis. Methods RFB from Wistar rats were isolated and cultured. Cell number was observed by MTT assay. Cell cycle was evaluated by flow cytometry (FCM). Results With the increase of urotensin Ⅱ concentration, the optical density at 490 nm of RFB as shown by MTF assay was first increased but then decreased, and remained at a significantly higher level in the cells treated with 1×10^-9 or 1×10^-8 mol/L UⅡ compared with those in control group(P 〈0. 05 ). With the treatment of 1×10^-10 ,1×10^-9 ,1 × 10^-3 ,1× 10^-7 mol/L U Ⅱ , the S phase cell percentage were significantly higher, and the GO-G1 phase cell percentage was significantly lower than those in control group(P 〈0. 01 ,P 〈0. 05), but the indices in 10-7 mol/L U Ⅱ group were not significantly different from those of control ( P 〉 0. 05 ). Conclusions U Ⅱ can directly induce RFB proliferation and affect the cell cycle, which suggests the crucial role of UⅡ in the renal interstitial fibrosis.
