摘 要:目的:建立柴胡中柴胡皂苷a、d含量的RP-HPLC测定方法,以分别考察不同种植基地柴胡及野生柴胡中柴胡皂苷a、d的含量。方法:采用BonChrom C_(18)色谱柱(200mm×4.6 nm,5μm),流动相为乙腈-水(40∶60),流速1.0ml·min^(-1),检测波长204nm。结果:柴胡皂苷a线性范围为1.94~19.44μg(r=0.999 9),柴胡皂苷d线性范围为1.96~19.60μg(r=0.999 8),柴胡皂苷a平均回收率为99.35%(RSD=1.14%),柴胡皂苷d平均回收率为99.52%(RSD=1.18%)。不同品种不同产地柴胡中柴胡皂苷a、d含量之和的分布范围为0.44%~0.67%。结论:本法简便快捷,结果准确,重复性好,地道中药材规范化种植(GAP)基地的柴胡皂苷含量较高。Objective: To establish an optimal RP-HPLC method to determine the content of saikosaponin a, d of Radix Bupleuri from different species or areas in China. Method: Saikosaponin a,d were separated on BonChrom ODS C18 eolumn(200mm ×4.6 mm, 5μm)with aeetonitrile-water(40: 60) as the mobile phase and with a flow rate of 1.0 ml ·min^-1. The detecting wavelength was at 204nm. Result: The linear ranges of salkosaponin a and saikosaponin d were 1.94 - 19. 44μg ( r = 0. 999 9 ) and 1.96 - 19.60 μg( r = 0.999 8), and the average recoveries of them were 99.35% (RSD = 1.14% ) and 99.52% (RSD = 1.18% ), respectively. The contents of saikasaponin a and saikosaponin d of Radix Bupleuri in different species or areas were 0.44% -0.67%. Conclusion: This method is simple, rapid, accurate and reproducible. Three GAP base areas of Bupleurum chinense have higher Saikosaponin contents.
