大豆凝集素在兔消化道不同部位内的免疫活性残留

摘  要:为揭示大豆凝集素在兔消化道不同部位的食糜中免疫活性的残留规律,用经N-乙酰基-D-半乳糖胺-epox-y-sepharose6B亲和层析体系纯化的大豆凝集素制备兔抗大豆凝集素多克隆抗体,建立大豆凝集素间接抑制ELISA定量检测方法,检测下限低于10ng.mL-1。以含质量分数为20%生大豆和1%Cr2O3(外源指示剂)的日粮饲喂未接触过大豆制品的兔(75日龄)14d,最后一次进食后2h,检测其胃、十二指肠、空肠前段、空肠中段、空肠后段、回肠、盲肠和结肠食糜中具免疫活性的大豆凝集素残留率分别为(74.33±2.57)%、(37.68±1.91)%、(12.15±1.97)%、(8.94±1.17)%、(7.34±1.45)%、(3.72±1.21)%、(2.04±1.19)%和(1.47±0.42)%。结果表明:在兔的消化道不同部位内均能检测到具有免疫活性的大豆凝集素,且由前向后总地呈下降的趋势,其中由胃到十二指肠和由十二指肠到空肠前段的残留率变化均达到了极显著水平(P<0.01)。To investigate the law of immune survival of soybean agglutinin (SBA)in different parts of digestive tracts in rabbits, SBA was purified with GalNac- epoxy- sepharose 6B affinity chromatography systems, and the polyclonal antibody of rabbit- anti- SBA were made. The way of quantitative determination of SBA with indirect inhibiting ELISA were established and the low limit was lower than 10 ng.mL^-1. Rabbits (aged 75d )were fed on the diet containing 20% raw soybean and 1% Cr2 O3 two weeks. Two hours after last meal, the survival SBA with immune activity of the thyme in stomach, duodenum, front-jejunum, middle-jejunum, behind-jejunum, ileum, appendix and colon were (74.33 ± 2.57 ) % , ( 37.68 ± 1.91 ) % , (12. 15 ± 1.97)% ,(8.94±1.17)% ,(7.34 ±1.45)% ,(3.72±1.21)% , (2.04 ±1.19)% and(1.47 ±0.42)% respectively. The results showed that there were SBA with immune activity in all of the parts of rabbits' digestive tracts, and it showed downtrend from stomach to colon. But the changes of the immune activity rate of SBA from stomach to duodenum and from duodenum to front-jejunum were all significant(P 〈 0.01 ).
