摘 要:目的观察恩诺沙星对鸡细胞色素P450 1A5(CYP1A5)表达的影响,为揭示兽药残留的产生机制以及临床安全联合用药提供理论依据。方法各实验组分别给予恩诺沙星5mg/kg(低剂量组)、25mg/kg(中剂量组)和125mg/kg(高剂量组);对照组给予相同体积的生理盐水。给药5d后检测鸡肝微粒体蛋白含量、CYP450含量、CYP1A5 mRNA及蛋白表达量的变化。结果中、高剂量组的肝微粒体蛋白含量、CYP450含量、CYP1A5 mRNA及蛋白表达量与对照组相比分别下降14%、36%,5%、5%,9%、20%,37%、49%,差异均有显著性(P<0.05)。结论中、高剂量恩诺沙星对CYP1A5的表达呈现出抑制作用。Objective To investigate the effect of enrofloxacin on the expression of cytochrome P450 1A5 (CYP1A5) in chickens, so as to provide theoretical basis for revealing the mechanism of drug residue formation, and clinical combined medication. Methods The treated chickens were given 5 mg/kg (low dose), 25 mg/kg (middle dose) and 125 mg/kg (high dose) of enrofloxacin while the controls were treated with the same volume of saline. Chicken hepatic microsomes were prepared by differential centrifugation. The concentration of chicken hepatic microsome protein was determined by the method of bicinchoninic acid (BCA). The content of CYP450 was determined by spectrophotometry. CYP1A5 mRNA was determined by real-time PCR and the expression of CYP1A5 protein was detected by Western-blotting. Results The concentration of hepatic microsome protein, the content of CYP450, the expression level of CYP1A5 mRNA and protein decreased 14%, 36%;5 %, 5 %;9%, 20%;37 %, 49% more respectively in both middle and high dose groups than in control group ( P 〈 0.05). Conclusion Middle and high dose of enrofloxacin has inhibitory action on the expression of CYP1A5 in chicken.
