摘 要:目的研究姜黄素固体分散体对四氯化碳所致小鼠急性肝损伤的保护作用。方法以聚乙烯吡咯烷酮(K30)为载体制备姜黄素固体分散体。将50只雄性昆明种小鼠随机分成5组:正常对照组、四氯化碳肝损伤模型组、姜黄组、姜黄素固体分散体低、高剂量组。采用腹腔注射四氯化碳致小鼠急性肝损伤模型,测定小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST);测定肝脏丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力;计算肝指数。结果与四氯化碳模型组比较,姜黄素固体分散体组小鼠血清中丙氨酸氨基转移酶和天冬氨酸氨基转移酶活力明显降低,姜黄素固体分散体能有效降低四氯化碳所致小鼠急性肝损伤,改善肝细胞损伤程度。结论姜黄素固体分散体对四氯化碳所致小鼠急性肝损伤有显著的保护作用。Objective To study the effects of cureumin solid dispersion on acute liver injury of mice induced by carbon tetrachloride ( CC14 ). Methods The curcumin solid dispersion was prepared with excipient of polyvinylpyrrolidone. Fifty mice were randomly divided into five groups : control group, carbon tetrachloride group, cureumin group, low and high dosage of curcumin solid dispersion groups. The acute liver injury model was established by intraperitoneal administered carbon tetrachloride. The contents of alanine aminotransferase ( ALT), asparate aminotransferase (AST) in serum and the activity or content of superoxide dismutase (SOD) and malondialdehtyde (MDA) in liver were measured to evaluate the protective effect of curcumin solid dispersion. Results The curcumin solid dispersion significantly decreased serum ALT activity, liver MDA content and SOD activity in CCl4 poisoned mice. Conclusion The curcumin solid dispersion protects mice against liver injury induced by CCl4.
