摘 要:目的以恩施州道地药材——五鹤续断为原料,初步研究其多糖提取分离及其动态分布规律。方法采用石油醚脱脂、热水浸提、乙醇沉淀分离、sevage脱蛋白、冷冻干燥等方法提取分离续断多糖,并采用蒽酮比色法测定续断多糖的含量,了解其多糖的动态分布规律。结果在提取2次、加料液比为1∶20、提取时间2 h、提取温度为95℃时,续断多糖得率最高,三年生续断根多糖分布最高。结论五鹤续断多糖分布规律为:三年生根>两年生根>一年生根>叶,人工种植的续断根>野生根。Objective To extract and separate polysaccharide in Dipsacus asper and extract polysaccharide and study its regularities. Methods During the extraction of polysaecharide, the effects of extraction times, temperature, proportion and time on yield of polysaecharide investigated by orthogonal experiment. The separation procedure of precipitation with alcohol and purification from the removing proteins and fats were analyzed. The contents of polysaecharides were determined by anthrone colorimetry. Results The results showed that the optimum parameters were as follows: 2 times extraction ratio of material solvent 1:20, the time 2h, the temperature was 95℃. Conclusion The regularities of polysaccharide distribution in Dipsacus asper is as follows: the polysaccharide in the roots grew for three years 〉 the one for two years 〉 for one year 〉 the one in the leaves, the cultivation Wu - He Dipsacus roots 〉 the wild form.
