高效液相色谱-蒸发光散射检测法测定毛冬青药材中Ilexgenin A的含量

摘　　要：目的建立毛冬青药材中Ilexgenin A的高效液相色谱-蒸发光散射检测（HPLC-ELSD）的含量测定方法。方法采用Lichrospher-C18色谱柱（4.6 mm×250 mm，5μm），流动相为乙腈-0.1%醋酸溶液（60∶40），流速1 ml/min，蒸发光散射检测器：漂移管温度96℃，气体流速2.1 L/min。结果Ilexgenin A在0.7-14.5μg浓度范围内线性关系良好，r=0.999 7（n=6）。平均回收率为100.32%，RSD=1.38%（n=6）。结论本法操作快速、准确，可作为毛冬青药材中IlexgeninA的质量控制方法。Objective To establish an HPLC - ELSD method for the determination of content of Ilexgenin A in Ilex Pubescens . Methods Liehroshper- C18 （4.6 mm × 250mm,5 μm） column was used in HPLC with mobile phase Acetonitrite -0.1% Acetic acid （60： 40）. The flow rate was 1ml/min, an evaporative light -scattering detector was used and the parameters were set as follows ： drift tube temperature was 96℃ and gas flow rate was 2.1 L/min . Results The linear range of Ilexgenin A is from 0.7 - 14.5 μg, r = 0.9997 （n = 6）. The average recovery is 100.23% ,RSD = 1.38% （n = 6）. Conclusion This method is rapid and accurate. It can be used for quality control on Ilexgenin A of Ilex Pubescens.
