摘 要:目的了解1.8 GHz移动电话射频电磁场(RF EMF)对大鼠神经细胞基因表达谱的影响,筛选射频电磁场反应基因。方法将原代培养的大鼠神经细随机分为实验组(辐照组)和对照组(假辐照组),经1.8 GHz射频电磁场,SAR为2 W.kg-1,间断辐照24 h(5 min开/10 min关)后,即刻抽提两组细胞的总RNA,用Affymetrix公司的Rat Neurobiology U34 array基因芯片筛选其差异表达基因,寻找可能的RF EMF反应候选基因。结果在1200个候选基因中,筛选出34个差异表达基因,其中上调基因24个,下调基因10个。差异表达基因与多种细胞功能相关(细胞骨架、信号通路、细胞代谢等)。这些基因的变化均小于2倍,但具有统计学意义(P<0.01)。结论1.8 GHz射频辐射影响大鼠神经元多个基因的表达,某些基因的改变提示射频辐射对神经元可能产生不良影响。Objective To investigate the changes of gnne expression in rat neurons induced by 1.8 GHz radiofrequency electromagnetic fields (RF EMF) and to screen RF EMF-responsive genes. Methods The rat primary cultured neuronal cells were divided into two groups, the radiation group and control group, from which the total RNA was extracted immediately and purified after intermittently (5min on/10min off) exposed or sham-exposed to a frequency of 1 .8GHz RF EMF for 24 hours at an average special absorption rate (SAR) of 2 W.kg^-1. Affymetrix Rat Neurebiology U34 array was applied to detect the changes of gene expression in rat neurons. Results Among 1200 candidate genes, 24 up-regulated and l0 down-regulated genes which are associated with multiple cellular functions (cyteskeleton, signal transduction pathway, metabolism, etc. ) after functional classification were found by using Affymetrix microarray suite software 5.0. Although the changes in gene expression were less than 2 folds, they had statistical significance (P 〈 0.01 ). Conclusion RF radiation of l .gGHz induce the changes of many genes transcription in rat neurons, Some of which indicate the negative effects of RF radiation on neurons.
