摘 要:目的研究BBP对大鼠肝脏细胞的作用。方法用不同浓度(5μmol/L,20μmol/L,80μmol/L)的BBP对大鼠肝脏细胞进行体外染毒1h,然后分别用硫代巴比妥酸(Thiobarbituric acid,TBA)法和KCl-SDS沉淀法来检测丙二醛(malondialde-hyde,MDA)含量和DPC系数的变化。结果各染毒浓度下MDA的含量较对照组均显著上升(P<0.01),并且具有很好的剂量-效应关系(R=0.973,P<0.05)。当BBP的浓度为5μmol/L和20μmol/L时,DPC系数与对照组相比略有上升但无显著性差异(P>0.05);当BBP的浓度上升为80μmol/L时,DPC系数明显上升,且与对照组相比有极显著差异(P<0.01)。结论BBP可对大鼠肝脏造成损伤,对机体有毒性作用。Objective To study the effects of BBP on rat hepatic cells. Methods Hepatic cells of rat were exposed to different con- centration of BBP (5 μmol/L, 20μmol/L and 801mol/L) for one hour in vitro. Thiobarbituric acid (TBA) assay and KCI -SDS assay were used to measure the change of the contents of malondialdehyde (MDA) and the coefficient of DNA - protein crosslinks (DPC) , re- spectively. Results That the contents of MDA in hepatic cells of all exposed groups obviously increased compared with the control group ( P 〈 0.01 ) and had a good dose - response relationship ( R = 0. 973, P 〈 0.05 ). DPC coefficient was increased but without significant difference (P 〉 0.05 ) when under the 5μmol/L and 20μmol/L of BBP. However, DPC coefficient obviously increased compared with the control group (P 〈0.01 ) when the concentration of BBP up to 80μmol/L. Conclnsiou BBP caused the injury to hepatic cells and had toxic effect on the organism.
