摘 要:目的研究三七总皂甙(Panax notoginseng saponins,PNS)对大鼠骨髓基质细胞(bone marrow strowal cells,BMSCs)分化为神经样细胞中神经生长因子(nerve growth factor,NGF)表达的影响。方法分离、纯化大鼠骨髓基质细胞(BMSCs),实验分对照组、诱导组、PNS组,分别采用常规培养液,常规培养液加入二甲亚砜(DMSO)、丁羟回醚(BHA)的诱导分化液及含100mg/LPNS的诱导分化液培养;观察细胞形态学变化,采用免疫组化及ELISA法了解细胞Nestin及NGF的表达。结果骨髓基质细胞体外诱导分化6h即出现形态学变化,表现为细胞胞体呈球形,突起增多,形态类似神经元。免疫组化显示Nestin染色阳性;诱导组、PNS组NGF表达明显低于对照组(P<0.01),PNS组高于诱导组(P<0.05~P<0.01)。结论骨髓基质细胞在体外可以定向诱导为神经细胞,诱导分化后NGF表达下降,PNS可以促进其神经诱导后NGF的表达。Objective To study the expression of NFG in neuronal differentiation of rat bone marrow stromal cells (BMSCs) in vitro. Methods BMSCs were isolated from femur and tibia of rats and cultured in basal medium(control), dimethylsulfoxide (DMSO) and butylated bydroxyanisole(BHA) in basal medium as induction group and containing PNS 100mg/L induction medium as PNS group. The differentiated BMSCs were detected by immunohistochemistry. The expression levels of NGF were determined by enzyme - linked im- munosorbent assay. Results During induction differentiation, Nestin were expressed by all BMSCs with ball -like shape with short process. The expression levels of NGF were inhibited during induction as compared with control(P 〈 0.01 ). The expression of NGF treatment PNS during induced was decreased as compared with control ( P 〈 0.01 ) , but increased as compared with induction group( P 〈 0.05 - P 〈 0.01 ). Conclusion BMSCs could be induced to differentiate into neuron - like cells in vitro. The expression of NGF was inhibited after induction and recovered by treatment by PNS.
