摘  要:目的研究三七总皂甙对大鼠骨髓基质细胞增生及向神经细胞分化的影响。方法实验将分离、纯化的大鼠骨髓基质细胞(bone marrow strowal cells,BMSCs)分为3组——对照组、诱导组和PNS组,分别采用常规培养液、常规培养液加入二甲亚砜(DMSO)和丁羟回醚(BHA)的诱导分化液以及含100mg/LPNS的诱导分化液进行培养,然后对各组细胞分别进行形态学观察,应用MTT法检测BMSCs增生及免疫组织化学方法检测BMSCs的Nestin表达。结果诱导6h后,少数BMSCs呈锥形,细胞突起增多伸长,类似神经元。继续诱导培养后细胞突起增多,突起相互连接成网状。诱导分化后细胞Nestin免疫染色阳性,PNS组阳性率>诱导组>对照组(P<0.05)。诱导组、对照组细胞增生良好,二者比较无显著差异(P>0.05),PNS组细胞增生明显,与对照组比较有显著性差异(P<0.05)。结论三七总皂甙可明显促进骨髓基质细胞向神经细胞的分化和增生。Objective To study the effects of total saponins of panax notoginseng on the proliferation of bone marrow strowal cells during differentiating into neuron - like Cells in vitro. Methods BMSCs were isolated from femur and tibia of rats and cultured in basal medium(Control), dimethylsulfoxide(DMSO) and butylated hydroxyanisole (BHA) in basal medium as induction group and containing PNS 100mg/L induction medium as PNS group. The morphological changes were observed under inverted microscope. The differentiated BMSCs were detected by immunohistoehemistry,and the proliferation were evaluated by MTT assay. Results During induction differentiation,nestin was expressed by all BMSCs with ball - like shape and short process. The nestin expression of PNS group increased as compared with induction group( P 〈 0.05 ). The proliferation of induction group had a slight increased compared with control group ( P 〈 0. 05 ). Conclusion The proliferation and differentiation of bone marrow strowal cells during differentiating into neuron - like Cells as facilitated by total saponins of panax notoginseng.
