重组马槟榔甜蛋白MabinlinⅡ在大肠杆菌中的表达

摘　　要：MabinlinⅡ是我国所特有且唯一的植物甜蛋白,在体外至今没有得到具有甜味的基因表达产物。本文根据已知马槟榔甜蛋白的序列设计引物克隆MabinlinⅡ基因,对基因进行剪切重组。将重组基因克隆至大肠杆菌表达载体pET-30a(+)中,构建了3个重组表达载体,转化大肠杆菌BL21(DE3),得到三株表达重组马槟榔甜蛋白的大肠杆菌工程菌。经IPTG诱导,3个重组MabinlinⅡ基因可在大肠杆菌BL21(DE3)中高效表达。Mabinlin Ⅱ is the unique plant sweet protein from Capparis masaikai which is only found in China but no sweetness gene expression products in vitro was reported so far. The recombinant Mabinlin Ⅱ genes were subcloned into a E. coli expression vector pET-30a（＋）. Three recombined plasmid pET-30a（＋） were then transformed into E. coli BL21 （DE3） host cells. Upon induction with IPTG, the recombinant Mabinlin Ⅱ protein were highly produced measured by SDS-PAGE.
