摘  要:目的:通过对比分析大负荷运动训练与模拟失重对大鼠下丘脑单胺类神经递质的影响,探究这两种应激源对机体HPG轴和HPA轴作用的中枢机制。方法:50只雄性SD大鼠随机分为安静对照组(CON)、游泳训练组(EXE)、后肢悬垂(模拟失重模型)组(HLS)、游泳训练补充睾酮组(ETR)和后肢悬垂补充睾酮组(HTR)5组;EXE和ETR大鼠进行6周递增负荷游泳运动训练,HLS和HTR大鼠在完成腹股沟管环缩手术后一周开始为期6周的尾部悬吊;ETR和HTR大鼠通过腹腔注射补充丙酸睾酮,剂量为2.5mg/kg体重/天,补充持续6周。EXE和ETR大鼠在完成最后一次游泳训练后即刻断头处死,CON、HLS和HTS大鼠在安静状态同时断头处死。大鼠断头后,迅速开颅取下丘脑,采用高效液相色谱—电化学方法检测单胺类神经递质。结果:EXE和ETR大鼠运动后即刻下丘脑单胺类神经递质DA、NE和5-HT相对CON大鼠的相应水平均有下降的变化趋势,但是(DA+NE)/5-HT有增加的变化趋势;HLS和HTR大鼠下丘脑单胺类神经递质DA、NE和5-HT相对CON大鼠的相应水平均增加,(DA+NE)/5-HT显著增加。结论:大鼠下丘脑单胺类神经递质和(DA+NE)/5-HT比值的变化可能是两种应激源导致机体HPG轴和HPA轴变化的中枢机制。Purpose. This study was to investigate the centre mechanism of the effects of heavy load exercise training and simulated microgravity on HPG axis and HPA axis in rats. Effects of the two stressors on monoamine neurotransmitters of hypothalamus in rats were compared. Method. Fifty male Sprague-Dawley rats were divided into gorups randomly as control group (CON), swimming training group (EXE), hindlimb suspension group (model of simulated microgarvity ) (HLS), swimming training group with supplementation of testosterone (ETR) and hindlimb suspension group with supplementation of testosterone (HTR). The EXE and ETR rats took a siex-week swimming protocol with training load increased progressively. The HLS and HTR rats endured a six-week suspension by tail one week after an operation of annulus abdominals shortened. Testosterone propinate was supplemented to the ETR and HTR rats by the way of intraperitoneal injection, with a dose of 2.5mg/kg body weight/day. The supplementation lasted for six weeks. The EXE and ETR rats were sacrificed by decapitation immediately after the last swimming training. The CON, HLS and HTR rats were decapitated at resting state at the same time. Hypothalamus was taken out as soon as the skulls of rats were opened after being decapitated. Monoamine neurotransmitters of hypothalamus were measured by electrochemical method with HPLC. It could be seen that the hypothalamus monoamine neuro-transmitters DA, NE and 5-HT of EXE and ETR rats immediate post-exercise tended to decrease when compared with those of CON rats, but the ratio of (DA+NE)/ 5-HT tended to increase. The monoamine neurotransmitters DA, NE and 5-HT increased significantly. Conclusion. It could be concluded that monoamine neruotransmitters and the ratio of (DA+NE) /5-HT changes of hypothalamus might be the centre mechanism for the changes of HPG axis and HPA axis induced by the two stressors in rats.
