摘　　要：以小型哺乳动物社鼠为例,通过调整酚与氯仿的比率、减少抽提次数及消化时间,对采用-70℃超低温冰箱、-20℃低温冰箱、70%乙醇液浸泡、95%乙醇液浸泡和含50 mmol/L乙二胺四乙酸二钠的70%乙醇液浸泡等不同方法保存的社鼠肌肉组织提取基因组DNA,并对其进行微卫星指纹图谱分析,研究了改进方法对不同方法保存的社鼠肌肉组织提取DNA的效果,并比较了不同保存方法对DNA提取效果的影响.结果表明:改进方法可行,完全可以满足微卫星等分子标记技术的需要.It was used the tissue of Niviventer confucianus by different preserving through adjusting the ratio of phenol and chloroform, reducing extraction and digesti preserved in different way including -70℃ refrigeration, -20℃ refrigeration cohol soak and 70% alcohol soak with 50 mmol/L EDTA. Microsatellite fingerp method to extract DNA, on time. The tissue were , 70% alcohol soak, 95% alrint was used to detect the extracted genome DNA in order to study the DNA extraction effect from the muscle tissue with different preserved way using improved method. It was approved that the improvement technique was feasible and effective. This method could be used with microsatellite and other molecular marker.
