消瘤汤对实验性肝癌大鼠肝脏血管内皮生长因子表达的影响

摘　　要：目的观察消瘤汤对二乙基亚硝胺诱导的大鼠肝癌模型肝脏血管内皮生长因子(VEGF)表达的影响,分析消瘤汤对肝癌的治疗作用及机制。方法清洁级雄性SD大鼠30只,随机分为3组:正常对照组10只,肝癌模型组10只,消瘤汤治疗组10只。模型组与治疗组大鼠给予10mg/kg二乙基亚硝胺(DEN,2g/L)灌胃,每周5次,间隔5h后,治疗组予消瘤汤2.5ml/100g(含生药3.6g/ml)灌胃,模型组灌服等量生理盐水;正常组于同样时间点给予等量生理盐水灌胃。于第16周处死各组大鼠,苏木精-伊红染色观察大鼠肝组织病理学结构的改变,免疫组化检测大鼠肝脏VEGF的表达。结果苏木精-伊红染色结果显示正常对照组大鼠肝小叶结构完整,肝细胞索排列整齐,细胞核清晰;模型组正常肝小叶结构被破坏,肿瘤细胞呈多形性,异形性明显,胞核增大,胞质少,嗜碱性;消瘤汤治疗组镜下癌细胞分化程度高,异型性低。免疫组化结果显示,与正常组VEGF(4.2±1.7)比较,模型组大鼠肝脏VEGF(75.3±11.5)呈弥漫性表达(P<0.01),治疗组大鼠肝脏VEGF(32.6±5.5)较模型组明显下降(P<0.01)。结论消瘤汤对实验性肝癌形成具有一定的抑制作用,其作用机制可能与下调肝肿瘤中VEGF的表达有关。Objective To investigate the effect of XiaoliuTang formula on the expression of vascular endothelial cell growth factor（VEGF）in rats with experimental hepatocarcinoma induced by diethylinitrosamine（DEN）. Methods Thirty male SD rats were randomly divided into 3 groups：control group（n = 10）, model group（n = 10） and traditional Chinese medicine（TCM）group（n= 10）. Except for the control group,the other groups were administered with 2 g/L DEN at a dose of 10 mg/kg（5 times/week for 16 weeks）. At the same time,the rats in the TCM groups were injected intraperitoneally with XiaoliuTang formula（9 g/100 g）once per day for 16 weeks. Hematoxylin and eosin（HE）staining were used to examine the changes of liver pathology and the expression of VEGF in liver tissues was detected by immunohistochemistry. Results Under microscope, the cancer cells in the rats of model group showed polymorphic and abnormal shape. In contrast, the cancer cells in the rats of TCM group were well differentiated. Immunohistochemistry observation showed that the expression of VEGF in the TCM group was remarkably decreased compared with the model group（P〈0.01）. Conclusion XiaoliuTang formula has inhibitory effect on the angiogenesis in rats with experimental hepatocarcinoma induced by DEN,which may be related to the downegulation of VEGF expression.
