摘 要:目的:研究拉伸应力下皮下疏松结缔组织内纤维和细胞形变及其对细胞活性的影响,探索中医替代疗法治疗机制。方法:实验组于SD大鼠腹股沟中点施加捻转拉伸,设置对照组。14d后制作皮下组织铺片、鬼笔环肽及碘化丙啶双染后相差显微镜下获取纤维形变图像、共聚焦激光扫描显微镜连续获取“细胞CT”图像、利用Mimics10.01进行细胞三维重建并通过BrdU掺入法评估细胞增殖活性。结果:在拉伸应力下,实验组纤维密度增高、排列呈与受力方向平行改变;细胞形态变为“扁梭形”、排列与纤维受力方向一致,对照组纤维和细胞分布无序;拉伸刺激能显著提高细胞的增殖活性。结论:拉伸应力下疏松结缔组织重构及对细胞增殖能力的影响可能是中医替代疗法的一种作用机制。Objective: To study the effects of tensile load on the structure and cell proliferative activity in loose connective tissue and then the mechanism of complement therapy was discussed. Methods: Tensile load exert on the midpoint of inguinal groove and control groups were set. 14 days later, stretched preparations were made and then observed under a phase con- trast microscope. After staining with phalloidin and PI, images were acquired by fluorescence microscopy and confocal laser scanning microscopy. At last, three-dimensional images were acquired from serial sections by mimics 10. O1 and cell proliferative activity was assessed by BrdU. Results: Under tensile load, the density of fiber and cells increased; The fiber and cells aligned themselves in the direction of tensile load. Tensile load can significantly increase the cell proliferative activity. Conclusion: Reconstruction of loose connective tissue and increasing of cell proliferative activity under tensile load may be one of the mechanism of complement therapy.
