摘 要:利用NCBI中公布的BAC序列信息开发新的SSR标记,在条纹叶枯病抗病基因Stv-bi区域附近设计了15对SSR引物,其中有3对引物(RM21-5、RM21-8、RM21-15)在抗病品种Achil106、关东194与不同感病亲本之间有多态性。采用Achil106×武育粳3号的F2∶3家系和7个抗、感组合的高世代稳定株系对这3个标记进行抗性选择效果评价。结果显示:3对引物对抗病型植株的分子检测与田间表型鉴定吻合率分别为80.8%、87.6%和82.8%;对感病型植株的分子检测与田间表型鉴定吻合率分别为92.4%、75.8%和88.6%。说明这些标记可以用来对相应组合的后代进行抗条纹叶枯病基因型的分子标记辅助选择。SSR markers were developed using the BAC sequences released at NCBI website. Fifteen pairs of gene-specific primers were designed at the region of Stv-bi, among which three pairs named RM21-5 ,RM21-8 and RM21-15 presented polymorphisms in the resistant varieties Achill06, Guandong 194 and some different susceptible parents. Resistance selection effects of three markers were further evaluated by using the F2:3 populations derived from cross Achill06 x Wuyujing No. 3 and 7 progenies derived from the crosses formed by resistant and susceptible varieties. It found that the coherence ratio to resistance type was 80. 8%, 87. 6% and 82. 8% between molecular identification and phenotypic data of fields experiment, and the coherence ratio to susceptible type was 92. 4%, 75. 8% and 88. 6%. It indicated that the molecular markers identified in this study could be helpful for marker-assisted selections of cross progenies for genotypes resistance to stripe disease.
