摘 要:苏玉20是江苏省农业科学院2004年选育成功的高产优质玉米杂交种。以苏玉20玉米杂交种及其亲本为材料,从80对玉米SSR引物中筛选出在苏玉20双亲之间多态性明显、重复性较好的4对引物bnlg1306、phi065、bnlg2291和umc1590用于苏玉20纯度鉴定。这4个SSR标记特异性好,且人为掺杂与田间鉴定均与利用SSR标记鉴定结果高度一致。尤其是,这些引物同时可以用于郑单958的纯度鉴定并可以区分苏玉20和郑单958杂交种。因此,该方法具有准确可靠、实用性强等优点,为生产上杂交种苏玉20纯度的快捷有效鉴定提供了科学依据。Maize hybrid Suyu 20 is a super cultivar with high yield and good quality bred by Jiangsu Academy of Agricultural Sciences in 2004. Genomie DNA extracted from Suyu 20 and its parents were subjected to SSR analysis with 80 pairs of SSR primers. As a resuh, four pairs of primers bnlgl306, phi065, bnlg2291 and umc1590 which could detect polymorphism between the parents were determined as candidates for purity assessment of Suyu 20. The purity test using the four specific markers was in agreement with the detection results of contrived fake seeds intermingling experiment and field identification. The four markers could detect hybrid seed purity of Zhengdan 958 and distinguish the maize hybrid Zhengdan 958 from Suyu 20 as well. Therefore, this procedure featuring accuracy and ease of use was applicable to test seed purity of Suyu 20 rapidly and efficiently using these SSR markers.
