摘  要:以苦瓜纯雌系X-黑-d-d为材料,于三叶一心期用300 mg/L AgNO3诱导其分化完全花,并利用cDNA-AFLP技术研究AgNO3处理后72 h内花蕾基因差异表达情况。结果表明:300 mg/L AgNO3能成功诱导苦瓜纯雌系连续性转化为完全花,平均为11.83朵/株;以128对选择性扩增引物扩增对照和处理后不同时期花蕾的cDNA,其中8对引物可获得较多分布均匀的清晰条带,这些条带大部分是组成型表达,少部分是抑制表达或诱导表达的差异条带。The transition of flower was observed after 300 mg/L AgNO3 application on gynoecious line X-Hei-d-d in bitter gourd at three-leaf and one apice stage, and the differential gene expression during bud development after the treatment in 72 h was studied by using cDNA-AFLP technology. The results indicated that 300 mg/L AgNO3 treatment could continuously induce female into complete flowers, in average 11.83 flowers per plant. Eight of 128 pairs of AFLP primer combinations could get evenly distributed and distinct fragments in the amplification of cDNA from the buds of the control and treatment at different time. Most of these fragments were composite expressive and others were depressive and up-regulatedly differential.
