摘  要:目的:建立宫颈癌的蛋白质组指纹诊断模型,筛选宫颈癌相关蛋白,为建立宫颈癌肿瘤标志物提供理论基础。方法:采用弱阳离子交换磁珠、基质辅助激光解析附电离飞行时间质谱(MB-WCX、MALDI-TOF-MS)技术对37例宫颈浸润癌(宫颈癌组)及50例健康人(对照组)的血清进行蛋白质组的对比研究。结果:宫颈癌组和对照组中共有21种蛋白质质峰谱强度差异有统计学意义(P<0.05),以M1450.35Da,M1778.7Da,M1896.65Da,M5520.42Da的4种蛋白质作为分类变量建立分类预测模型并对宫颈癌和对照组进行分类诊断,其识别率和预测能力分别为90.45%和81.75%;宫颈癌不同病理分化程度蛋白质组对比发现共有2种蛋白质质峰强度差异有统计学意义(P<0.05),以M5904.14Da,M5264.26Da的2种蛋白质作为分类变量建立分类模型并对不同分化程度进行分类诊断,其识别率和预测能力分别为81.48%和78.89%。结论:利用MALDI-TOF-MS技术筛选出来的具有分类意义的蛋白质,有可能成为宫颈癌诊断及预后判断的一组生物学指标,为宫颈癌肿瘤标志物提供重要线索。Objective: To establish a proteomic model for cervical cancer (CC) and to identify the biomarkers associated with CC through proteomic analysis of serum from CC patients and healthy controls. Methods: We applied MB-WCX and MALDI-TOF-MS methods to detect proteins in serum samples from CC patients (n= 37) and healthy controls (n=50) to find candidate proteins. Results: Twenty-one protein spectral peak intensities were significantly different between the CC patients and the healthy controls. Four proteins were identified, including M1450.35, M1778.7, M1896.65, and M5520.42, as classification variables for establishing the prediction model to identify and diagnose cervical cancer. The detection and prediction rates were 90.45% and 81.75, respectively. Two protein spectral peak intensities were significantly different among well, poor and moderately differentiated CC. Two proteins including M5904.14Da and M5264.26Da were identified as classification variables for differentiating CC. The detection rates were 81.48% and 78.89%, respectively. Conclusion: The proteins identified by our method may be related to CC and may be used as a group of biological markers in the diagnosis and prognosis of cervical cancer.
