摘  要:基于时间分辨荧光免疫分析和免疫层析技术的原理,研制了一种新型的时间分辨荧光免疫分析仪。与现有的时间分辨荧光分析仪不同,它所检测的是以Eu3+离子及其螯合物为标记物的免疫层析试纸条。通过测量免疫反应后试纸条检测带上Eu3+螯合物的含量,参照标准浓度曲线就可以定量得到待测样品溶液的浓度。实验表明,仪器的最低检测浓度可达到ng/mL量级,测量的相对标准偏差小于3%,在5~103ng/mL浓度范围内具有良好的线性响应特性,相关系数R2=0.9997。A novel time-resolved fluorometer based on immunochromatogragh er time-resolved fluorometers, it tests the immunochromatographie strip that y was developed. Different from the othis labeled with lanthanide ions and their chelates. This instrument can provide a rapid, quantitative measurement of analytes presented in the samples without any washing steps and it can be used to carry out point-of-care test (POCT). The instrument is composed of a specific optical sensor, a scanning stage, a signal processing system and a computer control system. A UV LED with pick wavelength of 345 nm is used as the exciting light source that is modulated at 1 kHz. The light from the LED is focused on the test strip by a focusing lens group. If the Eu3+ chelates are present in the samples, the fluorescence with pick wavelength of 613 nm will be exited. After a delay of some mLcroseconds, the fluorescence will be collected by the optical sensor and converted into electronic signal by a photomuhiplier tube (PMT). The concentration of the sample can be calculated according to the standard working curve of this instrument. Experiment test indicates that the mLnimum detection concentration is at several ng/mL level, the relative standard deviation is less than 3% and the test linear range is from 5 ng/mL to 103 ng/mL, within which the correlation coefficient R2 is 0. 999 7.
