摘  要:目的:研究CPNE5对TNF-α诱导的NF-κB转录激活活性的影响,并探讨其作用机制。方法:用10 ng/mlTNF-α处理转染了pcDNA3.1或pcDNA3.1-CPNE5的HEK293细胞,用双荧光素酶报告基因实验检测不同转染组NF-κB的转录激活活性;Western印迹和细胞免疫荧光技术检测CPNE5表达上调对NF-κB入核的影响;凝胶阻滞实验研究CPNE5表达上调对NF-κB DNA结合能力的影响。结果:CPNE5可以显著抑制TNF-α对NF-κB的转录激活活性(P<0.0001);CPNE5不影响TNF-α诱导的NF-κB(P65)入核;CPEN5过表达降低了TNF-α诱导的NF-κB与DNA结合的能力。结论:CPNE5通过抑制NF-κB的DNA结合活性抑制TNF-α诱导的NF-κB转录激活活性。Objective:To study the effect and the mechanism of CPNE5 on the transcriptional activity of NF-kB induced by TNF-α. Methods:HEK293 cells transiently transfected with pcDNA3.1 or pcDNA3.1-CPNE5 were treated with TNF-α. Dual-Luciferase Reporter Assay was used to study the transcriptional activity of NF-kB among different groups. Effects of CPNE5 on nuclear translocation of NF-kB were exemined by Western blotting and immunofluorescence staining. Effects of CPNE5 on DNA binding activity of NF-kB were studied by electrophoretic mobility shift Assay (EMSA). Results: CPNE5 inhibited transactivation activity of NF-KB induced by TNF-α ( P 〈 0. 0001 ). Although CPNE5 had no effect on nuclear translocation of NF-kB, CPNE5 inhibited DNA binding activity of NF-kB induced by TNF-α. Conclusion : CPNE5 represses transactivation activity of NF-kB by inhibiting its DNA binding activity.
