摘 要:目的探讨挫伤兔视网膜中血管内皮生长因子(vascular endothelial growth factor,VEGF)及表皮生长因子受体(epidermal growth factor receptor,EGFR)的变化。方法健康新西兰兔(普通级)40只,随机分为实验组(36只)和对照组(4只),以自由落体方式,用350g光滑铅球正面击打正常兔眼,观察对照组和实验组兔视网膜伤后不同时间点(伤后2h、1d、3d、7d、14d、20d)VEGF及EGFR的免疫组织化学变化情况。结果实验组挫伤兔视网膜中VEGF的表达先轻微下调,再随伤后时间延长而上调,伤后7d即高于对照组(P<0.05),20d时已回落至正常水平,即伤后7d灰度值为51.05±0.65,对照组为51.95±0.13(P<0.05);伤后20d实验组为51.03±0.69,对照组51.98±0.36(P=0.0503)。EGFR的表达在挫伤后2h、7d2组比较差异无统计学意义。结论挫伤后,兔视网膜中VEGF表达明显上调,而EGFR变化不大,提示一定强度眼挫伤后,组织细胞以局部修复为主,不出现大范围的细胞增生和分化,也不会产生广泛的新生血管形成。Objective To study the expressions of vascular endothelial growth factor(VEGF) and epidermal growth factor receptor(EGFR) in rabbit retina after ocular contusion. Methods Forty New Zealand rabbits at general class were randomly divided into experimental group (36 rabbits) and control group (4 rabbits). All rabbits were stricken by a freely falling copper ball, and immunohistochemical changes of VEGF and EGFR were observed at different times(2 hours, 1 day,3 days,7 days, 14 days and 20 days after contusion). Results After a slight drop, the VEGF showed a high expression in retina after contusion, and its expression was higher than that of control group at 7 days at contusion(P 〈0.05 ) ,returned to normal level at 20 days after contusion. At 7 days after contusion, the gray value of experimental group was 51.05 ± 0.65, and 51.95 ±0.13 in control group(P 〈0.05 ). At 20 days after contusion,the gray value of experimental group was 51.03 ± 0. 69, and 51. 98± 0. 36 in control group ( P = 0.050 3). The expression of EGFR had no statistical difference between the control group and experimental group at 2 hours and 7 days after contusion. Conelusions The high level expression of VEGF in retina after ocular contusion is meaningful. There is no significant change in the expression of EGFR. Present studies show these histiocytes with some abilities of tissue repair. There is no massive cellular proliferation, differentiation or neovascularization after contusion.
