摘 要:为了获得紫色大白菜细胞质不育系的特异序列,鉴定该不育系所属的不育类型,应用设计的orf138上、下游引物PCR扩增6份材料:紫色大白菜CMS不育系,保持系07-721,杂交F1,3个回交BC1单株BC1-1、BC1-2和BC1-3。对获得的差异片段进行克隆、测序,同源性比较。结果表明,在紫色大白菜CMS不育系、杂交F1、3个回交BC1单株上均扩增到309 bp的特异产物,而保持系07-721没有扩增产物。测序和同源性比对结果表明,紫色大白菜CMS的orf138与萝卜Ogura不育系的orf138、芸薹属作物与萝卜Ogu CMS体细胞杂种的orf138同源性均达到99%,E值为2e-155。证明了紫色大白菜CMS不育系为一种改良的Ogura萝卜细胞质不育系,同时初步探讨了该不育系和紫色基因的应用前景。In order to identify and certify the genotype of a purple Chinese cabbage CMS, and use the purple characteristics in Chinese cabbage, orf138 primers were used for PCR analysis in 6 materials including purple Chinese cabbage CMS, maintainer line 07-721, F1, 3 back-crossing segregating plants BC1-1, BC1-2, BC1-3. Then the PCR products were cloned, se- quenced and aligned. Results showed 309 bp fragments could only be amplified in purple Chinese cabbage CMS,F1 ,BCI-1, BC1-2, BC1-3, excluding maintainer line 07-721. Alignment results showed orf138 in purple Chinese cabbage CMS was homologous to Ogn CMS in Raphanus sativus and cybrids of Brassica crops with Raphanus sativus. The identities among them were 99% .E value was 2e-155. Therefore, the purple Chinese cabbage CMS material belongs to an improved Ogu CMS genotype. The application of the CMS genotype and purple genes in the future is discussed.
