摘 要:目的研究钾通道阻断剂四乙铵(TEA)对氧葡萄糖剥夺(OGD)诱导的培养海马神经元死亡的保护作用。方法培养12 d的海马神经元培养基更换为Earle′s平衡盐溶液(EBSS)后置于37℃三气缺氧(N2:CO2:O2=94%:5%:1%)培养箱内培养,4 h后恢复正常条件培养,同时在培养液内加入不同浓度钾通道阻断剂TEA,继续培养24 h后噻唑蓝(MTT)比色法检测神经元死亡情况。结果OGD组、OGD+不同浓度TEA组与对照组比较,海马神经元细胞存活率明显降低(P<0.05);OGD+10μmol.L-1TEA组和OGD+500μmol.L-1TEA组与OGD组比较,海马神经元细胞存活率明显升高(P<0.05);而OGD+1μmol.L-1TEA组和OGD+5 mmol.L-1TEA组与OGD组比较,海马神经元细胞存活率差异无统计学意义(P>0.05)。结论钾通道阻断剂TEA能保护OGD诱导的培养海马神经元免于死亡,提示某些类型的钾通道活动增强可能参与了OGD诱导的培养海马神经元死亡。Objective To investigate the effects of potassium channel antagonist on oxygen and glucose (OGD) -induced death of cultured hippocampal neurons. Methods After 12d's cultue,Earle's balanced salts(EBSS) took the place and then hippocampal neurons were put into the incubator with 37 ℃ and three anoxic gases( N2 : CO2 : O5 = 94% : 5% : 1% ). 4 h later culture under normal condition vitro was done again. Meanwhile, different concentrations of potassium channel antagonist tetraethyla mmonium(TEA) were applied to the culture solution. After 24 h, the neuronal death was analyzed with MTT assay. Results Compared with the control group,in OGD group and the group with different concentrations of OGD + TEA, the survival rate of hippocampal neurons was obviously lower(P 〈0.05) ; Compared with OGD group,in OGD + 10 μmol·L^-1 TEA group and OGD +500 μmol · L^-1 TEA group,the survival rate of hippocampal neurons was obviously higher(P 〈0.05); Compared with OGD group,in OGD + 1 μmol·L^-1 TEA group and OGD +5 mmol ·L^-1 TEA group,there was no statistical significance in the difference of the survival rate of hippocampal neurons (P 〉 0.05 ). Conclusion Potassium channel antagonist TEA can protect hippocampal neurons induced by oxygen and glucose deprivation from death, which indicates that enhanced activity of some kind potassium channel may get involved in the death of cultured hippocampal neurons induced by oxygen and glucose deprivation.
