摘 要:目的:了解日本血吸虫湖北株细胞色素C氧化酶亚基(mt co)基因的生物学特性.方法:以日本血吸虫湖北株成虫基因组总DNA为模板,设计特异性引物应用降落PCR扩增、纯化基因片段后连接、转化大肠杆菌DH5α,筛选阳性克隆、测序并与NCBI数据库中的核酸序列进行同源性比对分析,利用蛋白质分析软件分析该序列编码蛋白的相关生物学特性.结果:克隆序列的结果表明co基因全长1 581 bp,编码526个氨基酸;编码蛋白的等电点为6.28,相对分子量为59 KDα,具有一个细胞色素C氧化酶亚基的核心保守结构域.结论:日本血吸虫湖北株co基因与其他地理株具有较高的同源性.In order to explore the features of cytochrome c oxidase subunit Ⅰ (co Ⅰ ) gene from Schistosoma japonicum in Hubei strain,a pair of primers were designed and the template of adult Schistosoma japonicum total DNA from the strain was used to amplify a specific fragment by touchdown polymerase chain reaction which was purified and cloned into the E. coli DHSa. The inserts of the positive clones were identified first by double digestion, then by colony PCR and further sequenced. The data were analyzed with nucleotide BLAST software of NCBI,and the features of protein deduced from that sequence were analyzed with the protein database. Sequence analysis revealed that the ORF of co Ⅰ gene in S. j contained 1 581 nucleotides, encoding 526 amino acids and the molecular weight is about 59 kDa, PI is 6. 28. Structural predication showed this protein possessed a CO Ⅰconservative domain. It was concluded that the co Ⅰ gene of Schistosoma japonnicurn in Hubei strain had a high homology with the sequences from strain of other provinces.
