摘  要:目的观察NF-κB p65 siRNA对人子宫内膜异位种植于裸鼠的影响,以明确NF-κB在子宫内膜异位症(EM s)发生发展的作用,为EM s治疗探索新的途径。方法将EM s患者在位子宫内膜组织分别注入30只裸鼠,随机分为3组,每组10只,3 d后分别给予生理盐水、NF-κB p65 siRNA和阴性siRNA干预,观察各自子宫内膜种植成功率及内膜的病理特点。结果NF-κB p65 siRNA干预组子宫内膜种植成功率(20%)低于其他两组(80%,70%)(P<0.05);且成活的异位内膜腺体发育不良,组织中NF-κB、VEGF及ICAM-1表达亦减少,与其他两组差异有显著性(P<0.05);单纯接种组与阴性干预组间比较差异则无显著性(P>0.05)。结论NF-κB参与调控EM s的发生发展,沉默NF-κB基因可以显著抑制异位病灶的种植和生长;NF-κB是通过降调VEGF及ICAM-1等发挥作用的。Objective To investigate the processes regulated by NF -kB in the initial development of endometriotic lesions in vivo by means of siRNA targeting NF - kB interfering with the growth of endometrium. Methods Menstrual endometrium of women with endometriosis was injected into the peritoneal cavities of 30 nude mice. NF - kB p65 siRNA was given intraperitoneally on days 3 and 6, and mice were killed. The number, diameter and morphometry of endometriotic lesions were quantified. NF - kB, vascular epithelial growth factor (VEGF) and intercellular adhesion molecule (ICAM) - 1 expression were evaluated by immunohistochemical analyses. Results NF - kB p65 siRNA was significantly reduced in lesion development compared to control mice. Endometrial gland grew immaturely in treated mice. In addition, NF - kB activation, VEGF and ICAM - 1 expression of endometriotic lesions were significantly reduced. Conclusion NF - kB pathway is involved in the initial development of endometriosis in vivo, and may serve as a target for endometriosis treatment.
