1,4-二(2-异烟酰氨基-乙氨基)-5,8-二羟基蒽醌的合成与结构表征

摘　　要：以1,4,5,8-四羟基蒽醌隐色体和脂肪胺为原料,通过缩合、氧化、酰化反应合成抗癌候选化合物1,4-二(2-异烟酰氨基-乙氨基)-5,8-二羟基蒽醌(Ⅱ).考察了温度、溶剂、物料比等对产品收率的影响,确定较佳的工艺条件.对目标化合物利用IR,1H-NMR进行表征,产品收率60.0%.N, N＇-（ 2,2＇-（ 5,8-dihydroxy-9, 10-dioxo-9, 10-dihydroanthracene- 1,4-diyl ） bis （ azanediyl ） bis （ ethane-2, 1- diyl） ） diisonicotinamide （ Ⅱ ） as an intermediate of an anticancer drug was sythesized by the condensation, oxidation and acylation reaction of 1, 4,5,8-tetrahydroxy- 2,3-dihydroanthrace ne- 9,10-dione with aliphatic amine. The reaction conditions, such as the reaction temperature, the solvents and the materiel ratio were optimized. The structure of product was determined by FTIR and 1H-NMR analysis. The yield of compound Ⅱ was 60.0%.
