摘 要:目的研究痹肿消汤(Bizhongxiao decoction,BZXD)拆方(简称小复方)对小鼠体外T细胞活化、增殖及细胞因子基因表达的影响,探讨其免疫抑制作用及治疗类风湿关节炎(RA)的作用机制。方法无菌制备小鼠脾脏及淋巴结T淋巴细胞,加入不同浓度的小复方(0、12.5、25、50、100 mg/L),并用佛波醇酯和离子霉素刺激T细胞活化增殖;48 h后用MTT法检测T细胞增殖情况,6 h后用RT-PCR法检测白细胞介素-2(Interleukin-2,IL-2)mRNA和干扰素-γ(Interferon gamma,IFN-γ)mRNA表达。结果48 h后,PMA+Ion组细胞MTT吸光度(Ab-sorbance,A值)明显高于空白对照组,差异具有统计学意义(P<0.01);不同浓度的小复方组MTT吸光度均较PMA+Ion组减弱,差异具有统计学意义(P<0.01)。6 h后,PMA+Ion组IL-2mRNA和IFN-γmRNA表达均较空白对照组增高,差异具有统计学意义(P<0.01);不同浓度的小复方组的上述各细胞因子基因的表达与PMA+Ion组比较差异均具有统计学意义(P<0.01或P<0.05),呈不同程度的抑制。结论小复方对T细胞的活化、增殖及IL-2mRNA和IFN-γmRNA表达具有抑制作用,提示小复方具有免疫抑制及免疫调节作用,为痹肿消汤治疗RA的有效成分。Objective To study the effects of the separated portion (SP) from Bizhongxiao decoction (BZXD)on the activation,proliferation and cytokine of T lymphocytes and explore its immunosuppression and the mechanisms of it for rheumatoid arthritis (RA). Methods T lymphocytes of spleen and lymph nodes, prepared with asepticism, were added the BZXD in concentrations of 0, 12.5, 25, 50 and 100 mg/L and stimulated the activation and proliferation with PMA plus Ion; and T-cell proliferation was detected by MTT after 48 hours and the expression of IL-2mRNA and IFN-γ/mRNA was detected with RT-PCR after 6 hours. Results After 48 h,the absorbance in PMA plus Ion group was higher significantly than that in blank control group (P〈0.01); the absorbances in all SP groups were lower than the PMA plus Ion group (P〈0.01), After 6 hours of stimulation of PMA plus Ion,the expression of both IL-2 mRNA and IFN-γmRNA were in high level,and there were significant differences between PMA plus Ion group and blank control group (P〈0. 01). All the gene expressions of cytokine mentioned above in all SP groups were significantly different compared with PMA plus Ion group (P〈0.01 or P〈0.05), and inhibited in different degrees. Conclusions The SP has an obvious inhibition in the activation, proliferation and the expression of IL-2mRNA and IFN-γmRNA of T lym- phocytes, which shows that SP has the effects of immunosuppressive and immunomodulation and is effective constituent of BZXD for treatment of RA.
