摘 要:目的探讨愈痫灵颗粒(YXL)对痫性大鼠海马CA3区超微结构及认知功能障碍的影响。方法采用腹腔注射戊四氮点燃癫痫大鼠模型,随机分为癫痫模型组、愈痫灵颗粒干预组、丙戊酸钠干预组、拉莫三嗪干预组、正常对照组共5组;跳台实验测试大鼠的学习记忆能力,透射电镜观察海马CA3区超微结构。结果造模后反应期和学习成绩比较,正常组与其它各组差异有统计学意义(P<0.01),潜伏期和记忆成绩比较,正常组与其它各组差异无统计学意义(P>0.05);抗痫药物干预后各组反应期和潜伏期及学习、记忆成绩比较差异有显著统计学意义(P<0.001),愈痫灵组与正常组比较差异无统计学意义(P>0.05),愈痫灵组、拉莫三嗪组优于丙戊酸钠组与模型组,差异有统计学意义(P<0.05);抗痫药物干预前后反应期、潜伏期、学习、记忆成绩比较,愈痫灵组、拉莫三嗪组差异有统计学意义(P<0.01),丙戊酸钠组、模型组差异无统计学意义(P>0.05),正常组反应期、潜伏期、记忆成绩差异无统计学意义(P>0.05),学习成绩差异有统计学意义(P<0.01)。电镜观察模型组大鼠海马CA3区神经元可见明显损伤,愈痫灵组多数神经元结构正常,少数粗面内质网有轻度脱颗粒。结论愈痫灵颗粒减少戊四氮致痫大鼠海马组织神经元坏死可能是其改善癫痫大鼠认知功能障碍的作用机制之一。Objective To investigate the effect of Yuxianling granules (YXL)on epileptic hippocampus CA3 area ultrastructure and cognitive function. Methods The epilepsy rat models, induced by intraperitoneal injection of pentylenetetrazol, were randomly divided into model (model), Yuxianling granules intervention, valproate intervention, lamotrigine intervention, and control groups; the learning and memory ability of rats were tested with experimental platform, and the ultrastructure of hippocampal CA3 area with transmission eleetron microscopy observation. Results Compared with normal and other groups, the learning function in the reaction period after modeling was significant (P〈0.01), but in incubation and memory score was no signif-icant (P〉0.05); after anti-epileptic intervention in all the groups, the differences of reaction period and incubation, and learning and memory score were significant (P〈0.001), the difference between Yuxianling group and normal group was no significant (P〉0.05), in Yuxianling and lamotrigine groups, they were superior than that insodium valproate and model groups (P〈0.05); before and after the anti-epileptic drug intervention, the reaction period, the incubation period, learning and memory score in Yuxianling group were better than them in lamotrigine group (P〈0.01), the dif- ference between sodium valproate and model groups was no significant (P〉0.05), the normal reaction period, latency, memory score were no significant (P〉0.05), but learning score was significant (P〈0.01). With the electron microscope, hippocampal CA3 neurons was obviously injuryed in model group, the majority of neurons structure in Yuxianling group were normal, and minority of them in rough endoplasmic reticulum existed mild degranulation. Conclusions Lowering pentylenetetrazol kindling hippoeampus neuronal necrosis in epilepsy rats is one mechanisms of Yuxianling to improve the cognitive dysfunction.
