摘 要:目的:采用分子蒸馏技术对大豆油脱臭馏出物中维生素E进行纯化。方法:在其他条件固定的情况下,改变蒸馏温度,通过两级分子蒸馏纯化大豆油脱臭馏出物(未经任何化学处理)中维生素E;用RP-HPLC法作为维生索E各异构体定性、定量分析的手段。以邻苯二甲酸二辛酯为内标,用内标标准曲线法同时测定各馏分中维生素E各异构体的含量。结果:在系统压力为0.1 Pa、进料速率为1ml/min、刮膜速度为300r/min以及进料温度为50℃的条件下,一级、二级分子蒸馏的最佳温度分别为100℃和170℃;所得目标馏分中维生素E各异构体总含量由原料中的5.07%提高到29.76%。建立了同时测定纯化后馏分中维生素E各异构体含量的RP-HPLC定量分析方法,对所测定组分,平均回收率在93.1%~100.6%之间,相对标准偏差小于1.0%,线性相关系数大于0.999.方法检测限在(2.0~2.5)mg/L之间。结论:利用分子蒸馏技术可直接对未经酯化等化学处理的大豆油脱臭馏出物中维生素E进行纯化,方法简便、高效;所建立的反相高效液相色谱检测各馏分中维生素E的方法准确、可靠,对确定分子蒸馏纯化大豆油脱臭馏出物中维生素E的工艺条件具有指导意义。Objective: Molecular distillation technology was applied for extraction vitamin E from soybean oil deodorized distillate. Methods: Keep the other condition unaltered, vitamin E was refined from the soybean oil deodorized distillate (without any chemical pretreatment ) with quadratic molecular distillation under different distillation temperature. The qualitative and quantitative analysis of the isomers of vitamin E was conducted by means of RP - HPLC method. The contents of the isomers were simultaneously determined by internal standard curve method using dioctyl phthalate as internal standard. Results: When the system pressure, feed rate, rotor rate and feed temperature were 0. 1 Pa, 1 ml/min ,300 r/min and 50℃, the optimum temperature of the primary distillation and secondary distillation were 100℃ and 170℃ respectively. The yield of fractions of vitamin E was increased from 5.07% to 29.76%. A novel assay has been established for extraction vitamin E from soybean oil deodorized distillate by molecular distillation technology and determining the concentration of the isomers of vitamin E using RP - HPLC method sinmltaneously. The average added standard recovery was in the range of 93. 1 -100. 6 % and the relative standard deviations ( RSD ) were less than 0. 1%. The correlation coefficients were more than 0. 999 and the detection limits were in the range of ( 2.0 - 2. 5 ) mg/ L. Conduslons: Molecular distillation technology can be applied to refining vitamin E from soybean oil deodorized distillate without esterifying simply and efficiently. The RP - HPLC method can determine the concentration of vilamin E exactly and reliably. It has guiding significance for investigating the optimal process condilions of refining vitamin E from soybean oil deodorized distillate by molecular distillation.
