摘 要:目的:建立反相高效液相色谱法测定清热解毒方中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚含量的方法。方法:采用HPLC法,色谱柱:Kromasil C18(250 mm×4.6 mm,5μm),以甲醇-0.2%磷酸(85∶15)为流动相,流速:1.0 mL.min-1,检测波长:254 nm,柱温:35℃。结果:芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚分别在1.012 5~16.2μg、1.025~16.4μg、1.025~16.4μg、1.025~16.4μg、0.625~10μg范围内呈良好线性关系,相关系数分别为0.999 1,0.999 8,0.999 7,0.999 4,0.999 6,平均回收率分别为101.4%,102.0%,101.7%,102.1%,100.8%。结论:该方法操作简单、准确,且重复性好,可作为清热解毒方质量的控制指标之一。Objective: To develop an HPLC method for determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in QING RE JIE DU entraet. Methods: The analysis was carried out on a Kromasil C18 (250 mm × 4.6 mm, 5 μm)column eluted with a mobile phase of methanal-water containing 0.2% phosphoric acid solution(V:V = 85:15). The flow rate was 1.0 mL·min^-1 and column temperature was 35℃. The detection wavelength was set at 254 nm. Results: The calibration curve was linear within the range of 1.012 5 - 16.2,1.025 - 16.4,1.025 - 16.4,1.025 - 16.4 and 0. 625 - 10μg for aloe-emodin, rhein, emodin, chrysophanol and physcion, respectively. The average recoveries were 101.5 % ,99.2 %, 101.9 %, 101.5 %, 101.4 % for the abore five conponents, respectively. Conclusion: This method is convenient, accurate and reproducible for determination of aloe-emodin, rhein, emodin, chrysophanol and phascion in QING RE JIE DU extraet.
