摘 要:[目的]探讨高脂膳食诱导的肥胖及肥胖抵抗大鼠的血清肿瘤坏死因子α(TNF-α)、谷胱甘肽(GTH)及丙二醛(MDA)的水平。[方法]将雄性SD大鼠30只随机分为高脂组(21只)与正常对照组(9只),分别用高脂饲料和普通饲料饲养24周。根据高脂组大鼠体重增长程度,把高脂组大鼠分为肥胖组与肥胖抵抗组。采用酶联免疫法测定各组大鼠血清中TNF-α、GTH和MDA的含量。[结果]肥胖组大鼠的体重和脂肪总重量较正常对照组及肥胖抵抗组大鼠显著增加(P﹤0.05),肥胖抵抗组大鼠的体重与正常对照组比较无差异,脂肪总重量较正常对照组有增加趋势,但差异未达到统计学意义。肥胖组与肥胖抵抗组大鼠血清TNF-α水平较正常对照组显著升高(P﹤0.05),肥胖组与肥胖抵抗组两组间差异无统计学意义(P﹥0.05);肥胖组大鼠血清GTH水平较正常对照组显著降低(P﹤0.05),肥胖抵抗组大鼠血清GTH水平与正常对照组比较有降低趋势,但差异未达到统计学意义(P=0.059),肥胖组与肥胖抵抗组两组间差异无统计学意义;肥胖组与肥胖抵抗组大鼠血清MDA水平较正常对照组有升高趋势,但差异未达到统计学意义。[结论]肥胖抵抗组大鼠的体重与正常对照组大鼠比较无差异,但高脂膳食可能诱导了其体内炎症反应和氧化应激发生。[Objective] To observe the serum levers of tumor necrosis factor alpha (TNF-α), glutathion (GTH) and malonaldehvdc (MDA) in obesity-prone (OP) and obesity-resistant (OR) SD rats induced by high fat diet. [Methods] 25 male SD rats were randomly divided into high-fat diet group (n=21) and control group (n=9), and were fed with high-fat diet and normal diet for 24 weeks, respectively. The rats fed with high-fat diet were further divided into OP (n=7) and OR (n=7) subgroups, according to the body weight. The serum TNF-α, GTH and MDA levels were detemdned by ELISA. [ Results] Both body weight and total fat weight in OP group were significantly higher than those in OR and control groups, respectively, (P 〈 0.05). Ahbough the body-weight in OR group showed decreased tendency compared with that in control group, no significant difference was observed. Serum TNF-α levels in OP and OR groups were higher than those in control group, respectively, (P 〈 0.05), Mille there was no significant difference of them between OP and OR groups (P 〉 0.05). Serum GTH levels in OP group was lower (P 〈 0.05), whereas thcv were tended to be reduced in OR group (P=0.059), when compared with those of controls. respectively. There was no significant difference of them between OP and OR groups. Serum MDA levels in both OP and OR subgroups; were tended to be increased compared with those of controls (P 〉 0.05). [Conclusion] There is no significant difference in body weight of rats between OR and control groups, however, high fat diet could lead to in vivo inflammatory reaction and oxidative stress.
