摘 要:目的评估机械法、酶解法和酶解-机械结合法等分离方法对小鼠移植卵巢分离卵泡的发育及其生殖潜能的影响,建立更好的卵泡分离技术方案。方法B6D2F1新生小鼠卵巢移植后分别用机械法、长时酶解法、短时酶解法或酶解-机械结合法分离卵巢组织中的腔前卵泡,分离回收卵泡在体外培养12d时加入绒毛膜促性腺激素(hCG)处理26h,收获卵丘复合体进行体外受精。结果机械法的卵泡回收量、卵泡培养残存率、排卵率均高于酶解法,短时酶解法的排卵率高于长时酶解法;酶解-机械结合法的平均卵泡回收量、平均排卵数、卵母细胞成熟率等多项指标均高于其他各组。结论酶解-机械结合法是分离小鼠卵泡良好的技术方案。Objective To evaluate the effects of isolating methods (mechanical, enzymatical or the combined) on follicle development and its fertilizing potency in mouse ovarian graft. Methods Preantral follicles were isolated from mice ovarian graft by mechanical, long-term enzymatical, short-term enzymatical or meeh-enzym-combined method respectively. Follicles recovered were cultured in vitro for 12 days and hCG induced for 26 h. The cumulus masses were harvested and undergone with underwent IVF. Results The number of isolated follicles per ovary, survival rate in culture and ovulation rate of follicles were higher in mechanical method than those in enzymatical method. Ovulation rate was higher in shortterm than that in long-term enzymatical method. The number of isolated and ovulated follicles per ovary, the rates of maturation of oocytes were significantly higher in mech-enzym-combined method than those in other methods. Conclusion The mech-enzym-combined method is the best method ever evaluated.
