摘 要:目的探讨犬心房纤颤(atrial fibrillation,AF)模型中微囊蛋白-3(Caveolin-3)蛋白表达情况。方法建立犬房颤(AF)模型,通过免疫荧光染色和Western blot技术检测心房快速起搏建立的犬AF模型及AF患者心房肌的Caveolin-3蛋白表达情况。结果在犬和人房颤的心房组织,Western blot结果显示Caveolin-3蛋白表达明显降低,与非房颤组织相比P<0.05。在犬假手术组和窦性心律的患者,免疫荧光结果显示位于心房组织细胞膜上Caveolin-3蛋白表达明显,并均匀分布在细胞膜表面;而在AF状态下,人和犬心房肌Caveolin-3蛋白表达显著降低。结论Caveolin-3参与了AF这一临床复杂病症的发生和发展。Objective To investigate the expression of Caveolin-3 in canine model of atrial fibrillation (AF) and patients with AF. Methods Model of atrial fibrillation was estabhshed. Immunofluorescence and Western blot were used to determine the expression of Caveolin-3 in the tissues with and without AF. Results Both immunofluorescenee and Western blot showed the expression of Caveolin-3 was significantly decreased in AF tissues compared to that in the normal groups ( P 〈 0. 05 ). Immunofluoreseenee results of both canine group with sham-operation and patients with sinus rhythm showed that Caveolin-3 expressed obviously in the cell membrane of atria and distributed uniformly. However, expression of Caveolin-3 was significantly decreased in AF. Conclusion Caveolin-3 may be involved in the oceurance and development of AF.
