摘  要:应用ERIC—PCR法对鸭瘟病毒强毒株经皮下和口服感染20日龄鸭的十二指肠、空肠、回肠、盲肠和直肠中细菌菌群的结构多样性和动态变化进行了检测。结果表明:对照鸭肠道内的菌群结构相对稳定,其中十二指肠和空肠的ERIC—PCR条带最少,盲肠最多;皮下感染24h和口服48h前,各肠段ERIC—PCR扩增条带没有明显变化,皮下感染48h的鸭盲肠、口服感染72h的鸭盲肠和回肠条带数量明显增加,随着感染时间的延长,条带数呈逐渐减少的趋势,死亡鸭各肠段的条带最少。对ERIC—PCR扩增条带中主带的测序结果表明,大肠杆菌、志贺菌、沙门杆菌等需氧菌或兼性厌氧菌成为皮下感染鸭的直肠、口服感染鸭的盲肠(回肠和直肠)的优势菌群;口服感染鸭的回肠中一个未知菌成为优势菌群。An ERIC - PCR method was applied for analyzing intestinal microbial community diversity and dynamic changes of twenty - day - old ducks infected by hypodermic or oral routes with DPV virulent strain. The results indicated that the intestinal microbial community of controlled ducks was steady, and the band numbers of duodenal and jejunal ERIC - PCR were the least and the caeca/ were the most. The ERIC - PCR community profiles of orally infected ducks didn' t obviously changed until 24 h, while until 48 h in hypodermically infected ducks. Caecal ERIC - PCR bands of hypodermically infected ducks obviously increased at 48 h post inoculated and the same result was observed in ileum and caecum of orally infected ducks at 72 h post inoculated. ERIC - PCR bands of hypodermically and orally infected ducks gradually decreased with the extension of infected time and it were the least of dead ducks. The DNASTAR of staple of ERIC - PCR showed that aerobe and faeuhative aerobe ( Escherichia coli, Shigella, Salmonella ) had became preponderant bacilli in intestine of hypodermic and orally infected ducks. An unknown bacillus had became preponderant bacilli in ileum of orally infected ducks.
