摘  要:目的:观察冠心宁注射液对大鼠心肌缺血再灌注损伤梗死面积以及心肌热休克蛋白70(HSP70)表达的影响,探讨冠心宁的心肌缺血再灌注损伤的保护作用。方法:45只雄性SD大鼠随机分成3组(每组n=15):对照组(Ⅰ组)、缺血再灌注组(Ⅱ组)、冠心宁治疗组(Ⅲ组),建立在体心肌缺血再灌注动物模型。每组5只动物再灌注40min后测定梗死面积,测定超氧化物歧化酶(SOD)活性和心肌丙二醛(MDA)含量;每组另外10只动物再灌注120min后观察心肌细胞HSP70蛋白表达。结果:再灌注前注射冠心宁注射液显著降低了再灌注心肌的梗死面积、MDA水平(P<0.01),增强了SOD活性(P<0.01)和HSP70的表达(P<0.01)。结论:冠心宁注射液可通过抗氧化作用、增加HSP70的表达发挥抗心肌缺血再灌注损伤,降低心肌梗死面积,对大鼠心肌有明显的保护作用。OBJECTIVE To investigate the effect of Guanxinning injection on the rats myocardial infract size,the expression of heat shock protein 70 and to study the protective effect of Guanxinning injection on myocardial ischemia reperfusion iniury in rats. METHODS 45 male SD rats were randomly divided into 3 groups (n = 15) : control group (group Ⅰ ), ischemia-reperfu- sion group (group Ⅱ ) and Guanxinning injection group(group Ⅲ ). Myocardial ischemia-reperfusion model were established by using rats. After 40min ischemia, the 5 rats heart were harvested for determination of myocardial infract size, Malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of each group. The expression of heat shock protein70(HSP70) were measured of the other rat after 120 min ischemia. RESULTS The infraet size in group Ⅲ was significantly smaller than that group Ⅱ (P〈0. 01 ). MDA content of myocardium was reduced significantly in group Ⅲ (P〈0. 01 ), but significantly increased in group Ⅱ (P〈0. 01);SOD activity and HSP70 exprssion of myocardium in group Ⅲ were higher than those of group Ⅱ (P〈 0. 01 ). CONCLUSION Guanxinning injection can protect myocardium from ischemia-reperfusion injury via the improvement of HSP70 expression and reduce the infract myocardium size.
