摘  要:目的研究JNK信号通路在同型半胱氨酸(Hcy)诱导人脐静脉内皮细胞(HUVECs)凋亡中的作用。方法在人脐静脉内皮细胞培养基中加入不同浓度Hcy培养24h,以及加入SP600125预处理再加入10.0mmol/L的Hcy作用2h。用TUNEL法检测HUVECs凋亡情况。用Western-Blot法检测不同组HUVECs表达P-JNK的蛋白含量。结果用0.3mmol/L的Hcy干预后,HUVECs凋亡和JNK表达开始高于对照组(均P〈0.01)。100mmol/L最显著(均P〈0.01),呈浓度依赖关系;而SP600125两种浓度对HUVECs凋亡和JNK表达均有抑制作用,其中10.0umol/L更显著(P〈0.01)。结论Hey能够诱导HUVECs凋亡和上调JNK的表达,而SP600125具有明显的抑制作用,这可能是其促进动脉粥样硬化形成的重要机制之一。Objective To evaluate JNK signal pathway in apoptosis of human umbilical venous endothelial cells (HUVECs) induced by homocysteine (Hcy). Methods The HUVECs were with treated with Hcy in different concentrations for 24 h ; when 10.0mmol/L Hcy was used HUVECs were pretreated with SP600125 for 2 h. Cell apoptosis was detected by TUNEL method and P-JNK was measured by western blot. Results TUNEL-positive cells and the expression of P-JNK of HUVECs after exposure to Hcy were higher than those of control group, with a dose-dependent manner (P〈0.01) ; and the effects of 10.0mmol/L Hey were the most significant. Pretreated with the cell apoptosis and the phosphorylation levels of JNK induced by 10.0mmol/L Hcy were reduced (P〈0.01), and when SP600125 concentration was 10.0 μmol/L the effect were most significant (P〈0.01). Conclusion Homocysteine can induce apoptosis of HUVECs in vitro and upregulate the expression of JNK, which indicates that Hcy may be associated with the pathogenesis of atherosclerosis.
