摘 要:目的探讨宝灵膏对蒙古沙鼠脑缺血再灌注损伤脑组织梗死面积和神经细胞凋亡的影响及该影响是否存在剂量依赖性。方法采用双侧颈总动脉阻断的方法建造沙鼠脑缺血再灌注损伤模型。沙鼠随机分为假手术组、模型组、宝灵膏高剂量组、宝灵膏中剂量组、宝灵膏低剂量组。采用TTC染色法测脑组织梗死面积,采用末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)检测脑组织凋亡神经细胞。结果宝灵膏高、中剂量组TTC染色法测脑组织梗死面积与模型组比较差异显著,与假手术组无统计学意义;低剂量组测得梗死面积与模型组、假手术组比较差异显著。宝灵膏高、中剂量组采用TUNEL法检测脑组织凋亡神经细胞数与模型组比较有差异,与假手术组相近;宝灵膏低剂量组测得凋亡神经细胞数与模型组、假手术组比较有差异。结论宝灵膏能减轻沙鼠脑缺血再灌注损伤后脑组织梗死面积,抑制脑神经细胞的凋亡,并且其作用具有剂量依赖性。Objective: To observe the effects of bowring paste(BP) on cerebral infarction size and neuronal apoptosis after cerebral ischemia reperfusion injury(IRI) in Mongolian gerbil and the influence whether or not shows a dose-depended relationship. Methods: The bilateral carotid artery occlusion method was used to establish gerbil cerebral ischemia reperfusion injury model. The model gerbils were divided into five groups randomly: sham group, model group, BP high-dose treatment group, BP medium-dose treatment group, BP low-dose treatment group. The TTC staining was used to measure brain infarction area, and the terminal deoxynudeotidyl transferase meditated dUTP-nick end labeling(TUNEL) was used to determine brain nerve cells apoptosis. Results: The brain infarction size and apoptosis brain nerve cells of BP high, medium-dose treatment group had significant differences with the model group, and were not significant with the sham group. The brain infarction size and apoptosis brain nerve cells of BP low-dose group had significant differences with model group, the sham group. Conclusion: BP can reduce the gerbil cerebral ischemia reperfusion injury cerebral infarction area, and inhibit brain cell apoptosis. It show a dose-dependent relationship.
