摘 要:目的观察黄芪对高糖高脂条件下对系膜细胞增殖及产生细胞外基质(ECM)的影响。方法人系膜细胞单独培养,分为对照组、甘露醇组、高糖高脂组、黄芪组,MTT比色法观察系膜细胞的增殖情况,ELISA法检测细胞上清液Fn、ColⅣ含量。结果高糖高脂组系膜细胞增殖较对照组增高,黄芪组的系膜细胞增殖较高糖高脂组减低;高糖高脂组上清液Fn和ColⅣ含量较对照组升高,黄芪组的ColⅣ、Fn含量较高糖高脂组减低。结论高糖高脂可刺激系膜细胞分泌Fn、ColⅣ增加,高糖高脂刺激的ECM分泌增加与系膜细胞增殖有关。黄芪可抑制高糖高脂刺激的系膜细胞增殖及ColⅣ、Fn分泌增加,具有肾脏保护作用。Objective: To study the effect of astragalus membranaceus on the hyperplasia and extracellular matrix of mesangial cells in high glucose and lysophosphatidylcholine(LPC). Methods: The model of mesangial cells of diabetic nephropathy was established and divided into 4 groups: control group, mannitol group, high glucose and LPC group, astragalus membranaceus group. MTT method was used to observe the hyperplasia of mesangial cell. The levels of collagen Ⅳ and fibrocentin were determined by enzyme linked immunosorbent assay in the culture media. Results: High glucose and LPC promote the hyperplasia of mesangial cell. The hyperplasia of mesangial cell of astragalus membranaceus group was lower than that of high glucose and LPC group. The level of collagen Ⅳ and fibrocentin of high glucose and LPC group was higher than that of control group. The levels of collagen Ⅳ and fibrocentin of astragalus membranaceus group was lower than that of high glucose and LPC group. Conclusion: High glucose and LPC can promote the hyperplasia and the increase of collagen Ⅳ and fibrocentin of mesangial cell. Astragalus membranaceus can inhibit the hyperplasia and increase of collagen Ⅳ and fibrocentin and has protective effect on the kidney.
