摘 要:目的探讨新福菌素对人鼻咽癌细胞株CNE-2的体外杀伤效应及作用机制。方法体外培养CNE-2细胞,应用MTT比色法观察新福菌素对CNE-2细胞增殖的抑制作用。倒置显微镜下观察实验组细胞的生长及结构变化,同时采用逆转录-聚合酶链反应技术(RT-PCR)检测Bax mRNA基因的表达。结果新福菌素在体外对CNE-2细胞有较强的增殖抑制作用,能诱导CNE-2细胞凋亡;同时进行Bax mRNA基因表达检测发现对照组CNE-2细胞Bax mRNA表达呈弱阳性;实验组CNE-2细胞Bax mRNA表达均呈阳性,且随着新福菌素浓度的增加而逐渐增强。结论新福菌素对CNE-2细胞有明显的杀伤效应,杀伤机制可能是诱导鼻咽癌细胞凋亡,并与凋亡相关基因Bax mRNA表达上调有关。Objective To determine the apoptotic effect and its mechanism of sinoactinomycin on the human CNE-2 cells of nasopharyngeal carcinoma in vitro. Methods CNE-2 cells were treated with sinoaclinomyein at different concentrations. The growth inhibition effect of sinoacfinomycin was determined by MTT assay. Apoptosis was evaluated under inverted microscope. Bax mRNA expression was detected by reverse transcriptase - polymerase chain reaction ( RT-PCR ) . Results Sinoactinomycin significantly inhibited the growth of the CNE-2 cells. The Bax mRNA level was significantly increased in sinoactinomycin treated cells compared to non- treated cells in a concentration dependant manner. Conclusion Sinoactinomycin inhibits the growth of CNE- 2 cells due to apoptosis induced by the up - regulatoin of Bax.
