摘  要:目的建立茯苓中茯苓酸含量的测定方法,比较9个不同产地茯苓中茯苓酸的含量,为评价不同产地茯苓的品质提供参考依据。方法采用反相高效液相色谱法(RP-HPLC),Kromasil100-5C18色谱柱(250mm×4.6mm,5μm),柱温30℃,流动相为乙腈-0.4%乙酸(80:20),流速1.0mL/min,检测波长242nm。结果茯苓酸在0.48~2.40μg范围内与其色谱峰面积呈良好线性关系,r=0.9997,平均回收率99.96%,RSD=1.85%。9个产地茯苓中茯苓酸的含量有所差异。结论该方法简便、专属、稳定,可用于茯苓药材中茯苓酸的含量测定。Objective To establish a method for determining the content of Pachymic acid in Poria cocos, and compare the content of Pachyrnic acid of Poria cocos from nine different areas, in order to provide a basis to evaluate the quality of Poria cocos from different areas. Methods RP-HPLC method was used on Kromasil 100-5 C18 column (4.6 mm×200 mm, 5 μm), with acetonitrile-0.4% ethanoic acid (80 : 20) as mobile phase, flow rate of 1.0 mL/min, column temperature at 30 ℃, and detection wavelength at 242 nm. Results Pachymic acid showed a fine linear relationship with the peak areas between 0.48-2.40 μg (r=0.999 7). The average recovery was 99.96% with RSD= 1.85%. There was a difference in the content of Pachymic acid in Poria cocos from nine areas. Conclusion The method is proved to be convinient, stable, and feasible for quality assessment of Pachymic acid which is the main effective content in Poria cocos.
