摘 要:目的:建立柱前衍生HPLC法测定白土茯苓多糖中单糖的组成。方法:以三氟乙酸(TFA)水解白土茯苓多糖,水解产物加入1-苯基-3-甲基-5-吡唑啉酮(PMP)进行衍生化,采用反相高效液相色谱法,测定白土茯苓多糖中单糖的衍生物。色谱柱:KromasilC18柱(250mm×4.6mm,5μm);流动相:100mmol/L醋酸铵缓冲液(pH=5.5)∶乙腈=77∶23(V/V);检测波长:245nm。结果:白土茯苓多糖中含有甘露糖、鼠李糖、葡萄糖、半乳糖和木糖,其摩尔比为66∶1∶7590∶452∶528。结论:该方法简便、准确,可以测定出白土茯苓多糖中单糖的组成。Objective:To establish precolumn derivation HPLC method to determine the monosaccharides in Heterosmilax japonica polysaccharide. Methods:Heterosmilax japonica sample was hydrolyzed with trifluoroacetic(TFA) and derivated by 1-phenyl-3methyl-5- pyrazolone(PMP). The monosaccharide composition of Heterosmilax japonica polysaccharide was carried out by reversed-phase technique on a Kromasil C18 column with a mobile phase composed of 100 mmol/L ammonium acetate buffer( pH5.5 )and acetonitrile in the ratio of 77: 23. The detection was carried out at 245 nm. Results:The Heterosmilax japonica polysaccharide composed of mannose, rhamnose, glucose, galactose and xylose with amolar ratio of 66:1 : 7590: 452: 528. Conclusion: The HPLC method with precolumn derivatization is appropriate for the analysis of monosaccharide composition of Heterosmilax japonica polysaccharide and the method is simple, quick and accurate.
