摘 要:目的比较柴胡、红花、川芎3种中药单体抗大鼠肝纤维化的疗效并探讨其作用机制。方法以生理盐水为对照,将3种中药用于大鼠肝纤维化模型和大鼠肝星状细胞(HSC)。观察肝组织形态变化;检测血透明质酸酶、Ⅳ型胶原含量;MTT法检测HSC细胞增殖抑制率;免疫组化法检测基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶组织抑制因子-1(TIMP-1),转化生长因子β1(TGFβ1),血小板衍生生长因子(PDGF)的表达。结果与对照组比较,中药处理组的HSC细胞增殖抑制率升高(P<0.05);血清透明质酸、Ⅳ型胶原含量减少(P<0.05);肝组织TIMP-1、TGFβ1及PDGF的表达均减少(P<0.05)。结论3种中药均能减缓实验性肝纤维化的发生。其机制可能是这些中药能下调TGFβ1及PDGF表达抑制HSC增殖活化,从而下调TIMP-1/MMP-1而促进细胞外基质降解,减少其沉积。Objective To study the anti - fibrotic effect of bupleurum chinense, carthamus tinctorius and szechwan lovage rhizome and their possible mechanisms. Methods The three Chinese traditional drugs were given to rats with hepatic fibrosis and hepatic stellate cells (HSC) culture in vitro. Natural sodium was administered or added as the control. The following indexes were examined including the degree of hepatic fibrosis, serum levels of hyaluronic acid (HA) and type IV collagen ( C Ⅳ ) , expressions of tissue inhibitors of matrix metalloproteinase - 1 ( TIMP - 1 ) , matrix metalloproteinase - 1 ( MMP - 1 ) , ransfonning growth factor β1 ( TGFβ1 ) and platelet - derived growth factor (PDGF) in liver tissues. HSC proliferation was evaluated by MTF assay. Results Compared with the control group, the serum level of HA and CⅣ decreased significantly ; expressions of TIMP - 1, TGFβ1 and PDGF decreased significantly in all drug - treated groups ( P 〈 0.05 ). Conclusion Bupleurum chinense, carthamus tinctorius and szechwan lovage rhizome inhibit hepatic fibrosis. They may play roles through decreasing TGFβ1 and PDGF expressions and the ratio of TIMP - 1 to MMP - 1 to inhibit HSC activation and proliferation to decrease extracellular matrix.
