摘  要:转录因子基因OsBTF3在水稻品种日本晴悬浮细胞中受白叶枯病菌(Xanthomonas oryzae pv.oryzae,Xoo)诱导表达。为了阐明OsBTF3在水稻叶组织中的表达特征,本研究利用RT-Q-PCR技术,对经3种亲和性病原菌[水稻白叶枯病菌(Xoo)、水稻条斑病菌(Xooc)和稻瘟病菌(Mg)]接种和4种信号分子[脱落酸(ABA)、水杨酸(SA)、茉莉酸甲酯(MeJA)、乙烯(ETH)]诱导处理的水稻叶片中OsBTF3的转录本进行了定量分析。结果表明,OsBTF3对Xoo、Xooc和Mg侵染的基因表达反应均显著地受到诱导,但反应速度和强度略有差异。而4种信号分子对OsBTF3表达的诱导作用差异较大,ABA诱导活性最强,MeJA和ETH次之,SA诱导作用不显著。因此,OsBTF3基因表达不仅具有病原菌Xoo、Xooc和Mg的诱导性,而且也具有信号分子MeJA、ETH和ABA的应答性。It had been previously shown that the OsBTF3 gene encoding the RNA polymerase B transcription factor 3 was up-regulated in the suspension-cultured cells of rice co-incubated with Xanthomonas oryzae pv. oryzae (Xoo), the bacterial blight pathogen of rice. To elucidate expression dynamics of OsBTF3 gene in planta, the transcripts in the leaves of 2-week-old rice seedlings inoculated with the virulent strains of X. oryzae pv. oryzae (Xoo) , X. oryzae pv. oryzicola (Xooc) and Magnaporthe grisea (Mg), or treated with the chemical signal molecules including salicylic acid (SA), ethane (ETH), methyl jasmonic acid (MeJA) and abscisic acid (ABA), respectively, were quantified by real-time PCR assay. The OsBTF3 gene expression response was observed to be significantly induced by bacterial and fungal infection with Xoo, Xooc and Mg during the susceptible interactions. However, the differential induction of OsBTF3 by the signal molecules was found. OsBTF3 transcript was increased drastically with ABA, MeJA and ETH, weakly with SA. The results indicated that rice OsBTF3 gene was differentially expressed in response to pathogen infection and signal molecules, and was potentially involved in plant disease susceptibility response.
